Magnetic Bead DNA Extraction of Bone Gene Groups
1, Carefully drill the clean bone and get 0.2g of cancellous substance, and put it into a 1.5ml EP tube. Put 300µL lysis buffer in. Put the tube into a constant temperature ( 65°C ) water tank to incubate 30---60 minutes.
2, Take the tube out and get 200µL supernatant. Put the supernatant into another new tube and add 300µL bind buffer and 10µL magnetic bead fluid in. point vibrate 15times to mix up well, leave it at room temperature 25 °C for 10 min, reverse it several times every 2----3 min.
3, Magnetic separation:hold the EP tube in the left hand, hold the rectangular magnet in the right hand and put it on the bottom side of the tube ( 2/3 from tube mouth),and leave it there for half a minute. The beads will be absorbed onto the wall of the tube, then suck all the liquid ( it must be fully-sucked). The magnetic frame inside the kit can also be used to do the separation.
4, Put the EP tube on a normal non-magnetic tube rack, add in 600µL washing buffer, point vibrate it 15 times to mix up well ( or suck the liquid slowly 15 times to mix up well), leave the tube still for 1 min, then get the EP tube to do the magnetic separation to absorb the beads onto the wall of tube, then suck all the liquid ( it must be fully-sucked).
5, Repeat Process 4 for one time
6, Add 50µL ~ 100µL eluting buffer into the EP tube, suck the liquid slowly for 15 times to mix up well, leave it in a constant temperature ( 65°C ) water tank for 10min. Slightly shake the EP tube several times every 2---3 minutes.
7, If not for instant use, store the EP tube at 2~8°C. For instant use, rectangular magnet or magnetic frame can be used to do the separation in order to absorb the beads onto the tube wall, then suck the liquid and transfer it into a new centrifuge tube to do further experiments.
Note: For the DNA extraction of teeth, this method can work as a reference.
For the DNA extraction of hair, it should be hair with follicles 0.5 cm to the hair root. Approximately 10mg , add 30µLPK in, according to need.
For animal tissues, they should be cut into 1mm particles.
